Platelet receptor proteolysis: a mechanism for downregulating platelet reactivity.
The platelet plasma membrane is literally at the cutting-edge of recent research into proteolytic regulation of the function and surface expression of platelet receptors, revealing new mechanisms for how the thrombotic propensity of platelets is controlled in health and disease. Extracellular proteolysis of receptors irreversibly inactivates receptor-mediated adhesion and signaling, as well as ...
متن کاملProteolysis of platelet cortactin by calpain.
Cortactin, a substrate of pp60(c-)src and a potent filamentous actin binding and cross-linking protein, is abundant in circulating platelets. After stimulation of platelet aggregation with collagen, cortactin undergoes a dramatic increase in tyrosine phosphorylation followed by a rapid degradation. The cleavage of platelet cortactin was detected in lysates prepared using either Triton-containin...
متن کاملProteolysis of myosin during platelet storage.
Physical properties of actomyosin from either fresh or stored platelets have been compared. Actomyosin obtained from platelets after 3 days of storage contained myosin that was 60-80% degraded to myosin rod. No myosin rod was detected in fresh platelets. The platelet myosin rod is similar to the rod produced by limited proteolysis of skeletal muscle myosin.
متن کاملRegulation of cell growth by redox-mediated extracellular proteolysis of platelet-derived growth factor receptor beta.
Redox-regulated processes are important elements in various cellular functions. Reducing agents, such as N-acetyl-l-cysteine (NAC), are known to regulate signal transduction and cell growth through their radical scavenging action. However, recent studies have shown that reactive oxygen species are not always involved in ligand-stimulated intracellular signaling. Here, we report a novel mechanis...
متن کاملProteasome proteolysis supports stimulated platelet function and thrombosis.
OBJECTIVE Proteasome inhibitors used in the treatment of hematologic cancers also reduce thrombosis. Whether the proteasome participates in platelet activation or function is unclear because little is known of the proteasome in these terminally differentiated cells. APPROACH AND RESULTS Platelets displayed all 3 primary proteasome protease activities, which MG132 and bortezomib (Velcade) inhi...
